JetPEITM转染试剂
该试剂是阳离子化的大分子多聚乙烯亚胺(PEI)及其衍生物,其与外源基因结合形成颗粒状,与细胞表面的蛋白多糖结合,通过细胞”内吞作用”进入细胞。PEI-复合体具有质子海绵的特性,能吸收溶酶体的H+,在复合体中形成酸性环境使核酸酶失活,保护DNA免受核酸酶的降解。进入细胞后,JetPEI复合体囊泡肿胀破裂,将DNA释放到细胞质中。
特点:
l 转染效率高,重复性好
l 可转染许多种细胞系,对粘附细胞和悬浮细胞同样有效
l 对原代细胞也具有很好的转染结果
l 也可以转染寡核苷酸
l 培养基可含或不含血清
l 极低的细胞毒性
l 方法简便,省时省力
基本步骤
1) JetPEI溶液2μl和质粒DNA 1μg混合5-30分钟。
2) 将JetPEI-DNA复合体溶液加入到细胞液中(可以在含血清的培养基中)。
3) 检测
N/P比率(影响转染效率的关键)
N/P比率是对JetPEI复合体的所带电荷的度量。它指的是DNA分子带负电的磷酸基团与JetPEI中N残基的摩尔比,是衡量核酸与转染试剂比例的参数。从理论上讲,JetPEI/DNA复合体的电中性为N/P=2-3,但实际操作时,N/P=5-10效果较好。
转染试剂的类型
1.JetPEI转染试剂
JetPEI是PolyPlus-transfection的主导产品,毒性低,对细胞不会造成伤害。它能把基因转入各种粘附或悬浮的细胞系中以及原代细胞。
2.In vivo-JetPEI转染试剂
通过静脉、大脑、腹膜以及气管等体内途径进行转染,目前已在小鼠、大鼠、鸭、豚鼠和猴等动物身上取得成功。
3.Tagged JetPEIs:细胞介导的转染试剂
Tagged JetPEIs是偶联有配体的PEI衍生物,可将基因转入特殊的细胞。
1) JetPEI-Gal & in vivo-JetPEI-Gal(肝细胞介导)
PEI衍生物,偶联有半乳糖。受体细胞:细胞表面表达半乳糖特异性膜血凝素的细胞,如肝细胞。
2) JetPEI-Man & in vivo-JetPEI-Man(树突状细胞和巨噬细胞)
PEI衍生物,偶联有甘露糖。受体细胞:细胞表面表达甘露糖特异性受体的细胞,如巨噬细胞和树突状细胞。
3) JetPEI-RGD(上皮细胞和内皮细胞)
PEI衍生物,偶联有RGD肽链。受体细胞:细胞表面表达整合素的细胞,如上皮细胞和内皮细胞。 4.Fluo JetPEI:荧光转染试剂
1) JetPEI- FluoF
PEI衍生物,偶联荧光素FluoF(激发波长490nm,发射波长520nm),通常用于胞内定位研究,适于荧光显微镜和流式细胞仪的检测。
2) JetPEI- FluoR
PEI衍生物,偶联四甲基罗丹明FluoR(激发波长555nm,发射波长580nm),常用于双色标记和协同定位的实验中,如用于绿色荧光蛋白或FITC标记的质粒的转染。
3) In vivo-JetPEI-FluoF & In vivo-JetPEI-FluoR
PolyPlus-transfection也提供体内转染的荧光试剂,常用于生物分布研究。
5.JetPEI-Biotin和in vivo-JetPEI-Biotin
PEI衍生物,偶联Biotin,通过链亲和素检测JetPEI/DNA在细胞内的定位和分布。

概要

JETPEI ®细胞-巨噬transfects 原发性巨噬细胞,神经胶质和树突细胞被认为难以转染。它包含结合到表达特定甘露糖的膜受体,如巨噬细胞的细胞的甘露糖缀合的线性聚乙烯亚胺。

JETPEI ® -巨噬细胞也是的细胞系如RAW 264.7转染试剂。

JETPEI ® -巨噬细胞是甘露糖轴承的线性聚乙烯亚胺设计用于增强表达在其细胞膜受体甘露糖细胞的转染。细胞定位是的甘露糖残基的特异性细胞表面受体的结合,导致DNA复合物的内化的结果。

PEI Transfection

PEI Transfection

 

 

Transfection:

 

  1. Split 293T cells one day before transfection in DMEM/10% FBS medium:
    1. 6 well dish: 0.5×106 cells
    2. 10cm dish: 4.0×106 cells
    3. 15cm dish: 9.0×106 cells

 

  1. Prior to transfection bring all reagents to room temperature.

 

  1. In a sterile tube dilute total plasmid DNA (ug) in serum-free DMEM w/o phenol red (volume of media is 10% of final volume in culture vessel). Use transgene: viral packaging (psPAX2):viral envelope (pMD2G) constructs at 4:2:1 DNA ratio

 

  1. 6 well dish: 200ul + 3 ug of total DNA
  2. 10cm dish: 1mL + 7-8 ug of total DNA
  3. 15cm dish: 2mL +  11-12 ug of total DNA

 

  1. Add PEI (1ug/uL) to the diluted DNA. Mix immediay by vortexing or pipeting. The volume of PEI used is based on a 3:1 ratio of PEI (ug):total DNA (ug).

 

  1. 6 well dish: 9ul of PEI(1ug/ul) = 9ug
  2. 10cm dish: 21ul of PEI (1ug/ul) = 21ug
  3. 15cm dish: 33ul of PEI(1ug/ul) = 33ug

 

  1. Incubate 15 minutes at RT
  2. Add DNA/PEI mixture to cells

 

  1. Harvest transfected cells and/or viral supernatant at 48 hours post-transfection

 

 

Reagents:

 

PEI (1ug/ul) – PEI is Polyethylenimine 25kD linear from Polysciences (cat# 23966-2). To make a stock solution:

  • Dissolve PEI in endotoxin-free dH2O that has been heated to ~80°C.
  • Let cool to room temperature.
  • Neutralize to pH 7.0, filter sterilize (0.22um), aliquot and store at -20°C; a working stock can be kept at 4°C.
