原生Psalmotoxin1（PcTx1）是一种40个氨基酸的毒素，zui初分离自南美洲狼蛛弓形猴的毒液。¹

PcTX1的特征在于异常的四联体Lys ²⁵ -Arg ²⁶ -Arg ²⁷ -Arg ²⁸，其可能在毒素分子的表面形成强阳性“斑块”，构成通道结合位点识别的强候选区域。PcTX1的分子支架可能类似于先前描述的锥形蜗牛和蜘蛛毒素^5,6，并且包括由三个二硫键网格化的三链反平行β-折叠结构。¹

PcTx1（IC ₅₀ = 0.9nM ），并且特异性阻断H ⁺阳离子通道的特定亚类，酸感应离子通道1a（ASIC1a），并且能够区分两个接头变体亚型，并且不阻断ASIC1b通道（仅在其N端序列上不同）。此外，PcTX1一旦该子单元与另一个家族成员（ASIC2a或ASIC3）相关联，就失去了阻止ASIC1a的能力。¹

ASICs在脑和脊髓神经元丰富，涉及疼痛感觉，缺血性脑卒中机械补偿，学习和记忆。^2,3 在急性和神经性疼痛模型中，ASIC1a与PcTx1的特异性阻断导致在阿片受体上游工作的镇痛作用。⁴

使用碘化形式的毒素，PcTX1位点的结合鉴定了ASIC1a细胞外环的富含半胱氨酸的结构域I和II（CRDI和CRDII）。通道的区域1和4之间的二硫桥支持在其中PcTx1作用的CRDI4-CRDII结构域与质子⁷的结合位点的后M1区之间的紧密关系。

Alomone Labs Psalmotoxin-1抑制在非洲爪蟾卵母细胞中表达的ASIC1a的阳离子电流。

膜电位保持在5mV，并使用TEVC连续记录全细胞电流。在生理溶液（箭头指示）中，通过快速（〜2秒）暴露于pH 5引发ASIC1a电流。在水平条所示的时间内，通过灌注将5nM，10nM和20nM Psalmotoxin-1（＃STP-200）引入浴中。20nM的Psalmotoxin-1导致了pH依赖性内向瞬变的*但可逆的抑制。

Alomone Labs is pleased to offer a highly specific antibody directed against an extracellular epitope of rat GluN2A. Anti-NMDA Receptor 2A (GluN2A) (extracellular) antibody (#AGC-002) can be used in western blot, immunoprecipitation, immunocytochemistry, and immunohistochemistry. The antibody recognizes an extracellular epitope and is thus ideal for detecting GluN2A in living cells. It has been designed to recognize NR2A from rat, mouse, and human samples.
