ISOPLANT II

◆原理

      ISOPLANTII 是能够在短时间内提出去植物、酵母、细菌的 DNA 的试剂盒。和常规的 ISOPLANT 相同，Solution 的主要成分都是氯苯，能够破坏细胞壁，在界面活性剂存在下可以溶解。植物组织和动物组织相比，植物组织含有大量的多糖类和多酚，有可能会在提取 DNA 的时候混入。这些物质会抑制限制酶反应和 PCR，使我们难以对 DNA 进行定量。ISOPLANT II 是经过改良的能够除去这些物质的阻碍效果的试剂盒，而且不仅从草本植物，从木本植物中也能提取出 DNA，得到的 DNA 能用于 PCR 和限制酶反应中。

纯化的人拓扑异构酶IIα（Top2a）过表达，并在SDS-PAGE上纯化至单一条带的均一性。拓扑异构酶II高度纯化，无核酸酶污染。

人拓扑异构酶IIα质量控制试验

核酸污染：通过检测线性kDNA和线性质粒DNA的形成来检测。在37°C（在top2a测定条件下，含或不含ATP）下，将1µg连环KDNA或超螺旋pUC19 DNA孵育4小时。

通过在抑制人类topo IIa活性的条件下测定超螺旋DNA的松弛来评估与topo I的交叉污染。将过量纯化的酶与pRYG DNA在37°C条件下，在不存在ATP（含或不含Mg++）的情况下孵育2小时。在这些条件下，不得检测到pRYG超螺旋DNA的松弛。

当SDS-PAGE凝胶过载纯蛋白时，可以看到170kDa的主带。最终馏分中没有180kDa拓朴II形式。（不同批次的蛋白质浓度不同，但通常在5-50μg/ml的范围内）人类拓扑异构酶IIα的供应量为2-10单位/ul（37°C时，1个单位将在30分钟内降解0.2μg KDNA）。注意，单位定义基于优选的DNA底物kDNA（动粒DNA）。我们建议将kDNA底物与这种酶一起使用，因为质粒DNA弛豫测定几乎不那么敏感。活性基于kDNA脱链（而非质粒DNA松弛）进行验证。

top2酶的最终部分来自FPLC柱，在以下缓冲液中：10%甘油、50mM Tris（pH 7.7）、1mM EDTA和EGTA、650mM NaCl。

人拓扑异构酶IIα测定条件和性能质控

使用在拓扑II反应缓冲液（50mM Tris-Cl，pH 8.0，120mM KCl，10mM MgCl2，0.5mM ATP，0.5mM二硫苏糖醇）中最终体积为20-30µl的动载体DNA（KDNA）底物和0.2µg KDNA进行链连接测定。用5µl（每20µl反应体积）的终止缓冲液（5%沙科西、0.0025%溴酚蓝、25%甘油）终止反应。反应产物在含有0.5ug/ml溴化乙锭的1%琼脂糖凝胶上分析。当凝胶运行时，用手持紫外光源可以容易地监测2.5kb DNA微环脱链产物的分辨率。

酶在干冰上运输，应在-70°C下储存。我们还建议在第一次解冻后等分酶（反复冷冻/解冻会导致活性丧失）；酶活性稳定1-2天（不更长）。

产品描述

胶原酶（Collagenase）是一种蛋白酶，一种肽链内切酶，能够特异性的识别Pro-X-Gly-Pro序列（该序列高频率出现在胶原中，很少发现于其他蛋白中）并切割该序列中性氨基酸（X）和甘氨酸（Gly）之间的肽键。许多蛋白酶都能水解单链且变性的胶原多肽，但胶原酶是一种可以降解具有三股超螺旋结构的天然胶原纤维的蛋白酶，这种胶原纤维广泛存在结缔组织内。

本品来源于溶组织梭菌（Clostridium histolyticum），是一种酶粗提物，不仅含有胶原酶（更准确的称法为梭菌蛋白酶A（clostridiopeptidase A）），能够降解天然胶原和网状纤维。还含有其他的一些蛋白酶、多糖酶、脂酶等，分别能够有效水解结缔组织和上皮组织细胞外基质内的其他蛋白，多糖和脂质，使得本品非常适用于组织消化。

目前商业化提供的细菌胶原酶主要根据胶原酶活性的差异，分为四种类型：胶原酶I型，II型，III型和IV型，在应用上有所偏向：

1）胶原酶I（Type I Collagenase）：含有比较均匀的各种酶活力（包括胶原酶、酪蛋白酶、梭菌蛋白酶、胰蛋白酶活性）。通常用作上皮细胞、肝、肺、脂肪和肾上腺组织细胞的制备；

2）胶原酶II（Type II Collagenase）：含有更高的梭菌蛋白酶活性，通常用于心脏、骨、肌肉、胸腺和软骨等组织来源细胞的制备；

3）胶原酶III（Type III Collagenase）：含有较低的蛋白酶活性，常用于乳腺细胞的制备；

4）胶原酶IV（Type IV Collagenase）：含有低胰酶活性，通常用于胰岛细胞的制备，或者需要维持受体完整性的细胞制备实验。

本品为II型胶原酶，≥125 U/mg solid，可用于心、甲状腺、唾液腺、肝、骨、软骨等组织和细胞的解离。

酶活力单位定义

在37℃，pH7.5 的条件下，5 h内水解胶原产生相当于1 µM L-亮氨酸的酶量定义为1个酶活力单位。

运输及保存条件

室温运输。4℃避光保存，2年有效。储存液-20℃避光冻存。

使用方法

1. 胶原酶储存液的配制

向每管100 mg的胶原酶中加入100 µL的含Ca2+、Mg2+的HBSS（Hank’s平衡盐溶液，含Ca2+、Mg2+），轻轻旋涡震荡使其充分溶解，制备成1g/mL（即1000×）的储存液。然后用低蛋白结合性的0.22 μm的滤膜过滤除菌，分装成小份量，然后于-20 °C避光冻存。

使用前于冰上解冻，避免反复冻融。其用于组织和细胞分散的常用浓度为：0.5-2.5 mg/mL，用于软骨消化的常用浓度为1-2 mg/mL，需要根据特定的实验条件或者参考相应的文献资料确定所需的最佳工作浓度。

2．组织的分离

1. 使用无菌手术刀或剪刀将组织切成3-4 mm大小的组织块；

2. 利用含Ca2+、Mg2+的HBSS洗涤组织块数次；

3. 加入足量的含Ca2+、Mg2+的HBSS，使其浸没组织块，并加入胶原酶至需要工作浓度；

4. 于37℃孵育4-18 h。消化时使用水平摇床以及用3 mM的CaCl2补充消化可以提高消化效率。

5. 已分散开的细胞可使用不锈钢或尼龙网筛筛得，收集备用。未全部解离的组织另外添加适量的新鲜胶原酶工作液于37℃继续孵育；

6. 利用不含胶原酶的HBSS洗涤收集的细胞数次；

7. 细胞培养液重悬上述细胞，利用自动细胞计数器或其他方法计算活细胞密度。

8. 于细胞培养皿上利用合适细胞培养基接种细胞。

3． 器官灌注

1. 向37℃预热的含Ca2+、Mg2+的HBSS中加入胶原酶，另添加3 mM的CaCl2有助于提高分离效率；

2. 按照已优化的速率对相应的器官灌注胶原酶工作液；

3. 将上述过程中回收的灌注液流经不锈钢或尼龙网筛，从而将已解离的细胞或小片段组织块与较大团块分离开来，未充分解离的组织需利用新鲜胶原酶工作液于37℃进一步孵育；

4. 利用不含胶原酶的HBSS洗涤收集的细胞数次；

5. 细胞培养液重悬上述细胞，利用自动细胞计数器或其他方法计算活细胞密度。

6. 于细胞培养皿上利用合适细胞培养基接种细胞。

　　本试剂盒使用以邻苯三酚红(PR)为色素的比色法作为基本原理进行开发的。显色液操作简单，几乎不会对实验器皿造成污染。

◆测定原理


　　邻苯三酚红（PR）与钼酸结合，在 470 nm 处形成最大吸收波长的红色络合物。该络合物在酸性条件下与蛋白质结合，使波长发生移位，同时使试液呈现出蓝紫色。通过测定 600 nm 附近的吸光度，对试剂中总蛋白的浓度进行测定。

LC3 是一种 16-18 kDa 的小蛋白质，在细胞内以 3 种形式存在。125 个氨基酸的前体被切割成 120 个氨基酸的形式，称为 LC3-I。与磷脂酰乙醇胺 (PE) 结合后，生成 LC3-II 形式。LC3-I 主要存在于胞质溶胶中，而 LC3-II 主要存在于膜部分中。

注：可溶于酸性缓冲液（最大4mg/ml），但不易溶于中性缓冲液。为了将本品溶解于中性缓冲液中，首先将胶原蛋白溶解于0.01M或0.05M醋酸中，温度为1-4 mg/ml，然后将该胶原蛋白溶液添加到所需的中性缓冲液中，如2-10X TrisNaCl（NaCl最终浓度：0.15-0.2M），或在4°C下对1X中性缓冲液进行透析。

注1：为了避免在中性条件下形成纤维，将溶液放在冰上。

注2：II型胶原的理化性质不同于I型胶原和II型胶原

II在中性缓冲液中溶解的胶原蛋白在37°C培养时不会形成稳定的凝胶为了制备II型胶原蛋白凝胶，需要添加I型胶原蛋白或其他成分可能需要物质。
