现代Target Discovery的挑战需要复杂检测的自动化。免疫沉淀(IP)分析已被证明在研究发现中非常有用,以鉴定分子复合物的组分,发现蛋白质 – 蛋白质和蛋白质 – 核酸的相互作用,并量化免疫原性应答。

蛋白APhyTip®色谱柱已成功用于此目的,但是只有当目标肽或蛋白质具有抗体时,IP才能成功。生物素标记试剂的可用性允许研究人员通过将感兴趣的蛋白质缀合到抗生蛋白链菌素树脂上来生成定制的亲和树脂。这些定制树脂随即准备从生物样品中取出相互作用的蛋白质和复合物。为了有效地自动化这些分析,必须处理重要的样品制备步骤。也就是说,蛋白质需要被纯化,必须去除过量的生物素标记试剂,生物素化的蛋白质需要被固定。

PhyNexus亲和纯化,含5K分子量截留树脂的凝胶过滤柱和链霉亲和素柱联合使用,是小批量,自动化,高通量纯化,分离和固定生物素化蛋白用于复杂测定的理想工具。

 

 

 

 

 

 

 

 

PhyTip色谱柱是PhyNexus*的捕获,纯化和富集工具,专为实验室规模的蛋白质样品制备而设计。PhyTip色谱柱适用于各种液体处理平台,并含有特定的亲和树脂以满足特定的应用要求。

PhyNexus建议使用生物素标记的牛血清白蛋白(Sigma,A8549)作为标准来验证PhyTip色谱柱在所有应用中的使用。

纯化和富集的样品必须清晰,无颗粒物质。强烈建议离心样品,并使用PhyTip色谱柱前仅使用清澈的上清液。

 

链霉抗生物素蛋白PhyTip柱和链霉亲和素的PhyTip色谱柱已经过优化,可用于一般分子生物学实验室试剂和特定的PhyNexus仪器流速/体积,如下所示。这些信息是使用PhyNexus纯化系统收集的。

PhyNexus建议使用以下缓冲区(不提供):

PhyNexus建议使用含有IgG的PhyTip入门试剂盒作为标准来验证PhyTip色谱柱在所有应用中的使用。

纯化和富集的样品必须清晰,无颗粒物质。强烈建议离心样品,并使用PhyTip色谱柱前仅使用清澈的上清液。

运输和存储

每包PhyTip色谱柱已经制造并按照zui高标准进行了质量控制,并运送到保持盒中,以保持每个PhyTip色谱柱内特定亲和树脂的完整性。本产品在环境温度下运输,但收到时应存放在标准的实验室冰箱中,温度在4℃到8℃之间。

  • 不要冻结或冻结。

蛋白APhyTip®色谱柱的性能特征和分析步骤

介绍

PhyNexusPhyTip®色谱柱是创新性的纯化工具,从根本上简化了各种来源的蛋白质的捕获,纯化和富集。这些净化工具成功的关键是设计保留亲和树脂床的机制,具有zui小的死体积和zui大的捕获潜能。一组PhyTipcolumn结合蛋白A在特定的*条件下特异性结合来自不同来源的抗体(IgG),因此可以去除核酸和其他污染物。快速洗涤步骤后,纯化的抗体很容易用常用的低pH缓冲液洗脱。该技术允许特异性的高产量的IgG,取决于各种条件,并且提供在非常高浓度的载体蛋白如BSA存在下的高度选择性纯化。PhyTip色谱柱具有*的结合能力,可以结合大于100μg的IgG,并且可以有效地回收少至200 ng的IgG – 代表几乎三个数量级的范围。PhyTip色谱柱非常适合从杂交瘤上清液进行抗体筛选,或用于高通量提取和纯化亲和标记蛋白质。

绩效和分析程序

IgG以高亲和力和特异性结合蛋白A. 这种相互作用的强度和选择性使蛋白质A能够有效地从复杂的蛋白质混合物中纯化出IgG。为了检查用蛋白A树脂的PhyTip柱的性能,测量了使用1000个蛋白A PhyTip柱的纯化IgG(抗FITC-MAb)的回收百分比。

材料和方法

样品:在0.5ml 
含有5mg BSA的PBS中的10μgmFITC-MAb(IgG2a亚类)或
2.含有10%FBS(胎牛血清)的DMEM或
3.含有5mg BSA的SFM(无血清培养基)
样品处理:使用以下方案在半自动ME 1000平台上处理所有样品:

  1. (1)捕获:通过使样品在树脂床上以2个输入/输出循环(以0.6ml @ 0.25ml / min的体积设定)捕获特定蛋白质。
  2. (2)纯化:通过用0.5mL PBS(洗涤缓冲液I)洗涤结合的蛋白质/亲和树脂,使用1进/出循环(体积设定为0.6ml,0.5ml / min)除去未结合的蛋白质,随后是1进/出循环(体积程序为0.6ml,0.5ml / min)和0.5ml盐水溶液(洗涤缓冲液II)。
  3. (3)富集:用15μLpH 3.0洗脱缓冲液以4次输入/输出循环洗脱IgG2a(体积程序为0.1-0.15ml @ 1ml / min)。一旦洗脱,加入5μL中和缓冲液。请注意,PhyNexus提供的蛋白A和蛋白G PhyTip色谱柱的当前洗脱缓冲液的pH值为2.5,目前的中和缓冲液的pH值为9.0。

定量程序

(1)为了制备还原的IgG2a,将15μlzui终洗脱体积与5μl新鲜制备的TCEP(15μl10mg / ml TCEP的水溶液,终体积30μl)反应。

(2)使用HP 1050HPLC系统将20μl还原的IgG2a注射到无孔聚苯乙烯二乙烯基苯反相(C-18)柱中。溶剂A(0.1%TFA的水溶液)和溶剂B(0.1%TFA的ACN溶液)之间的梯度为25%至75%,使用5分钟。检测:在214和280nm处的UV。

  1. (3)大约3.18和3.34分钟时洗脱主要的IgG2a峰。在这两个峰下面的区域在每种情况下从(3.13-3.5)分钟积分,相应的峰面积在214nm处记录。
  2. (4)在相同的反应条件下将TCEP处理的IgG2a标准物加载到柱子中并用作回收计算的输入或标准。

IgG2a恢复的结果

在DMEM + 10%FBS中的回收率=在SFM + 5mg BSA中的77%回收率= 64%。

已知载体蛋白如白蛋白诱导免疫应答并有助于产生良好的抗体,所以当抗体产生时其通常以高量存在。因此,任何纯化和富集程序必须能够耐受高浓度的白蛋白。另外,上面显示的结果似乎表明高背景蛋白质浓度(例如BSA)的存在可能对回收率具有有益的影响。

载体蛋白存在下IgG2a回收率的重现性

为了验证载体蛋白在zui大IgG2a回收率中的作用,并证实重现性,从多个样品(n = 4)中从PBS(以及含有5mg BSA的PBS)纯化IgG2a。结果表明在存在500倍过量的BSA浓度的情况下提高了纯化的IgG2a的恢复(至少20%或更多),并且该程序也是高度可重现的。

在PBS中回收= 42.25%(SD = 4.66%)在PBS中回收+ 5mg BSA = 66.5%(SD = 7.5%)

选择性性能和分析程序

为了表征和验证蛋白A-IgG亲和相互作用的选择性,将5mg BSA作为载体蛋白加入到0.5ml样品体积(333倍过量)中的15μgIgG中。使用蛋白A PhyTip柱处理样品。

材料和方法

样品:在含有5mg BSA(10mg / ml或1%w / v BSA)的0.5ml PBS或PBS中的15μgmFITC-MAb(IgG2a亚类)。样品处理:如上所述的恢复性能和分析程序。

IgG定量和纯度分析程序:
