Kit Contents:

Solid CCI-103F (Hypoxyprobe™-F6)

Diluted Rabbit anti-CCI-103F antisera (PAbF6)

Applications: Use the Hypoxyprobe™-F6 Kit to measure hypoxia in tissues and cells by means of immunochemical detection including immunoperoxidase, immunofluorescence or flow cytometry. (Kleiter et al., Int. J. Radiation Oncol. Biol. Phys. 64(2): 592-602, 2006). Ljungkvist et al., Int. J. Radiat. Oncol. Biol. Phys. 62(4): 1157-1168, 2005).

Quantities: a. Hypoxyprobe™-F6 Kits contain 100, 200 or 1000 mg, respectively, of CCI-103F (Hypoxyprobe™–F6). Typical doses are 60mg/kg body weight is used in small animal studies.

b. Each Kit contains diluted anti-CCI-103F rabbit antiserum (PAbF6) containing 0.09% sodium azide and 1% BSA as stabilizers. The 100 and 200 mg Kits contain one 200 uL unit of antiserum; the 1000 mg Kit contains two 200 uL units of antiserum. Optimal operating dilutions of the antisera are to be determined by investigators but 1/200 -1/500 dilutions give strong immunoperoxidase staining when combined with peroxidase conjugated goat anti-rabbit secondary antisera.

Not supplied: Anti-rabbit secondary reagents or other immunohistochemical reagents (buffers, antigen retrieval agents, etc).

Storage: a. Store Hypoxyprobe™–F6 in the dark at room temperature (can also be stored at 2-8 degrees C)

b. Store rabbit antisera (PAbF6) at 2-8 degrees C.

Kit contents: Solid pimonidazole HCl (Hypoxyprobe™-1) and affinity purified rabbit anti-                               pimonidazole antibody (PAb2627AP)(ca 0.5 mg/mL IgG)

Applications: Immunochemical detection of cell and tissue hypoxia through immunofluorescence                      or immunoperoxidase staining, western blotting, or flow cytometry. Very low                               background in mouse tissues when the Hypoxyprobe™-1 Omni Kit is combined                         with an anti-rabbit secondary reagent (secondary reagent not supplied in the                               Omni Kit).

Quantities:     a.  Hypoxyprobe™-1 Omni Kits contain100 mg, 200 mg or 1000 mg of                                    Hypoxyprobe™- 1 (pimonidazole HCl). Typical doses are 60mg/kg body weight                        for small animal studies and 14mg/kg body weight for human studies

        b. One vial (100 and 200 mg Kits) or two vials (1000 mg Kit) of 200 microliters                         of affinity purified anti-pimonidazole rabbit antisera containing 0.09% sodium                               azide and 1% BSA as stabilizers. Optimal dilution of PAb2627AP is to be                                  determined by each investigator but a 1/50 -1/200 dilution has been found to give                       strong immunostaining in mouse tumor tissue when combined with peroxidase                              conjugated goat anti-rabbit antisera.

Not supplied: Secondary anti-rabbit reagent and standard IHC reagents such as buffers,                                  substrate chromogens, etc.

Storage: a. Store Hypoxyprobe™-1 solid at room temperature in the dark (can also be stored                  at 2- 8 degrees C).

              b. Store rabbit PAb2627AP at 2-8 degrees C. Do not freeze

Hypoxyprobe™ Plus Kit

Kit contents:

Solid pimonidazole HCl (Hypoxyprobe™-1)

FITC conjugated to anti-pimonidazole mouse IgG1 monoclonal antibody (FITC-MAb1) and

Rabbit anti-FITC conjugated with horseradish peroxidase as a secondary reagent.

Applications: Immunochemical detection of cell and tissue hypoxia through immunofluorescence or immunoperoxidase staining, western blotting, or flow cytometry. There is very low background in mouse tissues when the Hypoxyprobe™ Plus Kit is combined with an anti-FITC secondary reagent.

Quantities: a. Hypoxyprobe™ Plus Kits contain100 mg, 200 mg or 1000 mg of pimonidazole HCl. Typical doses are 60mg/kg body weight in small animal studies and 14mg/kg body weight for human studies.  b. One vial (100 and 200 mg Kits) or two vials (1000 mg Kit) containing 200 uL of an anti-pimonidazole, FITC-conjugated IgG1 mouse monoclonal antibody (clone 4.3.11.3). Each vial contains 200 microliters of a 0.50 mg/ml solution of the FITC-MAb1 in PBS containing 1 % BSA and 0.09% sodium azide. Optimal dilution of the FITC-MAb1 is to be determined by the investigator but 1:50 or 1:100 dilutions have been found to give strong immunostaining in mouse tumor tissue when combined with a 1:50 or 1:100 dilution of the peroxidase conjugated anti-FITC secondary reagent. While not designed specifically for immunofluorescence, FITC-MAb1 can be used for this purpose.  Antibody dilution is typically lower for immunofluorescence than for immunoperoxidase detection.

Not supplied: Standard reagents used for immunohistochemical analyses.

Storage: a. Store Hypoxyprobe™-1 solid at room temperature in the dark (can also be stored at 2- 8 degrees C).  b. Store FITC-MAb1 and HRP conjugated rabbit anti-FITC at 2-8 degrees C in the dark.    Do not freeze.

Solid pimonidazole HCl (Hypoxyprobe™-1) 

Anti-pimonidazole mouse IgG1 monoclonal antibody (MAb1) (60 micrograms/ml)

Applications: Immunochemical detection of cell and tissue hypoxia through immunofluorescence or immunoperoxidase staining, western blotting, or flow cytometry. 

Quantities: a. Hypoxyprobe™ Kit contains 100 mg, 200 mg or 1000 mg of pimonidazole HCl. Typical doses are 60mg/kg body weight for small animal studies and 14mg/kg body weight for human studies b. 1.0 or 2.0 ml of exhausted supernatant from hybridoma clone 4.3.11.3 containing 60 micrograms/ml of an IgG1 mouse monoclonal antibody and 0.15% sodium azide. Optimal dilution of MAb1 is to be determined by investigators but a 1/50 dilution has been found to give strong immunostaining in mouse tumor tissue when combined with a secondary strategy comprised of biotin-conjugated F(ab’)2 antimouse secondary reagent, streptavidin peroxidase and peroxidase substrate. Note that alternative secondary strategies can be used.

Not supplied: Secondary reagents such as biotin conjugated F(ab’)2 antimouse secondary reagent, streptavidin peroxidase or standard IHC reagents including buffers, peroxidase substrate, etc.

Storage: a. Store pimonidazole HCl solid at room temperature in the dark.

      b. Store the mouse monoclonal antibody (MAb1) frozen. Avoid repeated freeze thaw cycles by aliquoting antibody into small volumes for storage at -20 degrees C.
