3D细胞培养

如今,大部分细胞在塑料培养皿的表面生长,也就是说,是二维的。但在体内,细胞则被其他细胞、组织和细胞外基质重重包围。 

    于是,研究人员开始尝试3D细胞培养,并观察它与2D培养的差异,包括基因表达、信号转导、细胞增殖、细胞形态、细胞间相互作用以及药物反应的改变。

    幸运的是,从2D到3D细胞培养系统的转换并非想象中那么困难,而且也是相当值得的。“你的收获在于所研究的细胞更接近它们的真实环境,这对你的生物学研究有好处,”TAP Biosystems的RAFT开发主管Grant Cameron如是说。

    下面我们就来介绍一些从2D向3D转换时的注意事项。

3D系统的类型

    3D细胞培养基质(或支架)为细胞培养环境增添了第三维。它们有几种主要的类型,包括来自天然材料的水凝胶支架,如细胞外基质胶原蛋白。TAP Biosystems的RAFT(Real Architecture for 3D Tissue)技术正是基于胶原蛋白,它让研究人员能够开展细胞生物学、药物开发和组织工程等研究。

    Life Technologies的Geltrex®和BD Biosciences的BD Matrigel™是从小鼠肿瘤中提取出的基底膜基质,Sigma-Aldrich的MaxGel™是人细胞外基质,而Life Technologies的AlgiMatrix来源于褐色海藻。Microtissues公司在其3D 培养皿上使用基于琼脂糖的水凝胶。据介绍,在这种培养皿上,细胞会聚集并自我组装成数百个大小均一的多细胞球体。

    中所使用的第二种类型的支架是合成的水凝胶,有时也包括仿生肽,它们可增强整合素的结合。BD Biosciences的BD™ PuraMatrix™肽段水凝胶和Sigma-Aldrich的HydroMatrix™就属于这一类型,而Cellendes的3D Life Biomimetic 水凝胶也是由聚合物和交联剂制成的。

    其他类型的人造支架是来自组织培养的塑料,甚至是可生物降解的塑料。还有一个系统,是利用磁悬浮让细胞悬浮,其中细胞产生自己的细胞外环境,而无需塑料支架。

    n3D Biosciences公司的Bio-Assembler™系统就是这种,利用磁悬浮来3D培养细胞。该公司的科学家Glauco Souza介绍:“我们利用磁性纳米颗粒来实现,而无需支架。你可以利用磁性笔来移动细胞,创建组织层,然后拿走磁铁,细胞就自由了。”Souza领导的研究小组利用这种方法生成了一种四层的细胞结构,类似于肺组织。

    此外,还有一些系统可诱导球体形成,如B-Bridge的Lipidure-Coat板、3D Biomatrix的Perfecta3D®悬滴板,以及STEMCELL的AggreWell™,适合人诱导多能干细胞和胚胎干细胞。

何时转换

    越来越多的证据表明,在3D培养系统中生长的细胞会表现出更生理的表型、相互作用及反应。因此,当结果的生理相关性很重要,或者你无法通过其他方式来准确重现细胞行为时,或许应该考虑向3D转换。

    AMSBIO的业务部Peter Rettenberger认为:“细胞在平坦的塑料表面上生长,这与它们天然的生理环境相距甚远,也需要一定程度的适应。与之相关的是对细胞的某种选择压力。”

    当然,对于了解报告基因表达的转染实验而言,3D培养可能并不重要。但如果你研究的是复杂的药物相互作用,情况就大不相同了。在3D培养中,细胞对抗癌药物的敏感性可能与活体肿瘤更为相似。3D微组织中的药物运输和扩散更像3D的器官和组织。

    现在你准备好了?以下这些秘诀可帮助你平稳过渡。首先,注意接种密度,需要比2D培养更高。Cameron解释道:“许多细胞要彼此靠近,甚至要与其他细胞类型共同培养。每孔中的细胞数量应增加,10-20倍是个合理的考虑因素。”他还建议在开展实验之前让细胞有一些过渡时间。

    选择的3D支架材料也是成功过渡的一个关键因素。Cardy建议考虑几个关键问题:“我是否需要模拟细胞的天然细胞外基质?是否需要细胞形成有功能的多细胞结构?是否需要通过显微镜来分析我的3D培养,和/或回收细胞用于进一步分析?我是否想要控制凝胶的刚度和生物活性成分?”这些问题的答案可以帮助你缩小选择范围。

小心陷阱

    支架的选择也为3D培养的过渡带来陷阱,因为它让一些分析方法复杂化。显微镜观察就变得很有挑战性。2D培养的单层细胞很容易成像,而3D培养物的多层细胞会散射光线。共聚焦显微镜则很好地解决了这个问题,但其他类型的分析也许还很难。Cardy认为,许多支架都不适合免疫组化技术或细胞复苏。

    此外,细胞的行为也可能与2D有很大区别,例如,形成更的结构,如癌症肿瘤样。正如Cameron所言:“开辟了一系列新的细胞反应和可能,但这些也会让研究人员大吃一惊。”

VitroGel 3D的多种应用

1.我应该选择哪种产品,VitroGel 3D或VitroGel 3D-RGD?

如果您使用贴壁细胞系,建议您使用VitroGel 3D-RGD。如果您使用非贴壁细胞系,或者您想要更灵活地控制生长因子/蛋白质,我们建议您使用VitroGel 3D。请support@thewellbio.com如果你需要一个定制的产品。

2.产品有多稳定?

VitroGel 3D在4-40°C之间是稳定的。请勿将产品冷冻或将水凝胶溶液加热至80°C以上。该产品在室温下4℃或6个月未开封未开封18个月。一旦打开,请密封盖子,保持在4°C,以防止1个月内污染和使用。

3.如何调整水凝胶形成时间并作为可注射水凝胶进行准备?

水凝胶形成时间高度取决于水凝胶溶液和触发溶液(细胞培养基/PBS)的混合比例。我们建议将水凝胶溶液和介质/ PBS以4:1(v / v)的比例混合成可注射的水凝胶。水凝胶在混合后30分钟内应稳定。较高的混合比(水凝胶溶液:介质> 4:1)会导致更长的水凝胶形成时间。低混合比(水凝胶溶液:介质<1:1)可能导致不可注射的水凝胶。

4.水凝胶形成速度太快,我该怎么办?

水凝胶形成时间取决于几个因素,如凝胶浓度,培养基的离子强度和混合比。如果水凝胶形成太快,我们建议用DI水稀释水凝胶溶液,然后与细胞培养基混合使其凝胶化。稀释后,尝试保持4:1的比例(稀释的水凝胶溶液:细胞培养基= 4:1)进行混合,因为这是我们用于我们的水凝胶系统的大多数培养基的*比例。

5.当将凝胶溶液与细胞培养基混合时,我可以做什么来保持气泡形成?

气泡问题与将凝胶溶液与细胞培养基混合后溶液粘度增加有关。这里有一些有助于减少气泡形成的建议:
1)将水凝胶溶液预热至37°C以降低凝胶溶液的粘度。
2)将凝胶溶液和细胞培养基轻轻混合,并沿着孔板的壁缓慢移液,不引入气泡。(有些用户喜欢使用涡流而不是移液管进行混合以避免气泡)
3)在与细胞培养基混合之前用DI水稀释凝胶溶液。
4)切割移液器吸头以获得更好的流量。

6.如何调整zui终水凝胶的硬度?

通过在与细胞培养基/PBS混合之前稀释水凝胶溶液可以调节zui终水凝胶的硬度。请使用无菌去离子水稀释水凝胶溶液。不要使用PBS或其他离子溶液。如果您需要比原始产品更高的水凝胶刚度,请与我们。

7.应该使用什么种子密度?

我们建议对该水凝胶系统使用200,000-1,000,000个细胞/ mL。您可能需要相应地每1-2天更换一次覆盖的培养基。

8.细胞在VitroGel 3D水凝胶中生长多长时间?

我们已经测试了水凝胶系统中细胞的生长长达两周。依靠细胞系,3D细胞培养可以持续更长时间。一旦单元格的数量和大小增加,您可能需要更频繁地更换封面介质。
