肌动蛋白结合蛋白降速测定试剂盒提供G-或F-肌动蛋白加阳性（α – 肌动蛋白）和阴性（牛血清白蛋白，BSA）结合对照蛋白。肌动蛋白结合发生在对肌动蛋白的任何部位具有亲和力时。可以通过使用降速法测量F-肌动蛋白结合。以这种方式，离心用于通过差异沉降将F-肌动蛋白与G-肌动蛋白分离。F-肌动蛋白结合蛋白将与肌动蛋白丝共沉淀并在离心管底部形成沉淀。可以检测具有F-肌动蛋白成束活性的蛋白质，因为它们将导致F-肌动蛋白以比正常更低的离心力沉降（14,000xg对150,000xg）。F-肌动蛋白切断蛋白，G-肌动蛋白结合蛋白或非肌动蛋白结合蛋白将留在上清液中。如果上清液中残留的G-肌动蛋白比阴性对照样品中的G-肌动蛋白更多，则可以预期切断蛋白质，并且应该通过在添加测试蛋白质之前和之后测量F-肌动蛋白长度分布来进一步测试该活性。G-肌动蛋白结合蛋白可以通过将测试蛋白添加到G-肌动蛋白并诱导聚合来测量，如果测试蛋白螯合G-肌动蛋白，那么在旋转测定期间，与对照相比，肌动蛋白将留在上清液中。

问题1：当使用非肌肉肌动蛋白结合蛋白生物化学试剂盒（Cat。＃BK013或肌动蛋白结合蛋白生物化学试剂盒Cat。＃BK001）时，制备肌动蛋白结合试验蛋白的*条件是什么？

答案1：  在肌动蛋白相容缓冲液（如HEPES或磷酸盐缓冲液）中使测试蛋白质达到尽可能高的浓度。高浓度（优选>20μM）是优化检测肌动蛋白和测试蛋白之间相互作用的机会所必需的。在某些情况下，K d肌动蛋白结合蛋白/肌动蛋白相互作用的（解离常数）可以低到不能通过颗粒的蛋白质测定检测到; 然而，对于肌动蛋白在反应混合物中的影响可能足以通过沉淀和上清液中的肌动蛋白量与阴性对照的差异来检测。我们还建议将测试蛋白质在150,000 xg下在4°C下离心1小时，以去除变性蛋白质和细胞碎片。如果已经采取其他步骤（例如亲和柱纯化）来纯化测试蛋白，则这不是必需的。高速离心后，取出上清液置于冰上。这是测试蛋白质原料。

 

总离子强度应低于50 mM，这是由于肌动蛋白结合的特性，因此它主要是离子的，因此会受到中盐和高盐缓冲液的影响。一个好的起始缓冲液是20 mM Hepes pH 7.4加20 mM KCl，加上蛋白酶抑制剂，磷酸酶抑制剂和任何必要的辅助因子。可以用20mM MgCl 2和10mM ATP 制备低盐肌动蛋白聚合10x缓冲液，从而使盐在终反应中保持低水平。 

 

需要考虑的其他问题是（i）测试蛋白辅助因子可能是必需的，（ii）测定中的因子可能是主动抑制相互作用，（iii）测试蛋白可能偏爱不同类型的肌动蛋白，或（ iv）测试蛋白质在组织提取物中可以是低浓度的。

 

为了解决这些潜在问题中的一些，可以添加试剂以去除抑制剂，例如用于钙的EGTA（部件号BSEG-01）。在运动蛋白的情况下，ATP / Mg2 +组合将使运动与F-肌动蛋白解离，因此以1mM使用不可水解的类似物如AMPPNP。肌动蛋白亲和柱可用于浓缩和分离更多的测试蛋白。后，不同的pH条件可以改善肌动蛋白和肌动蛋白结合蛋白之间的结合（例如，α-辅肌动蛋白的适pH为7.0）。

 

问题2：非肌肉肌动蛋白结合蛋白生物化学试剂盒（Cat。＃BK013）可用于测定非肌肉肌动蛋白和肌动蛋白结合蛋白之间的结合亲和力吗？

答案2：   结合亲和力（K d）可以通过旋转降低测定来估计，通过滴定测试蛋白质浓度在0.2和20μM之间，并找到原始蛋白质在颗粒中的一半的浓度。绘制百分比测试蛋白质结合对比反应中的原始浓度以获得以μM计的Kd。

 

问题3：细胞裂解液可以与非肌肉肌动蛋白结合蛋白生物化学试剂盒（Cat。＃BK013）一起用作测试蛋白的来源吗？

答案3：是的，细胞裂解液可用作测试蛋白的来源。然而，细胞骨架不建议这样做，因为蛋白质的纯度和浓度通常太低而不能与肌动蛋白相互作用。此外，裂解物将含有额外的辅助蛋白和多种磷酸酶和蛋白酶，它们可以干扰或改变肌动蛋白和肌动蛋白结合蛋白之间的相互作用。如果要使用裂解物，我们建议如下：

产品用途包括

· 固定组织切片和组织培养细胞制剂中肌动蛋白丝的荧光染色。  注意：与许多肌动蛋白抗体不同，Acti-stain™488鬼笔环肽仅与F-肌动蛋白结合，导致低背景荧光。此外，通过Acti染色结合F-肌动蛋白在物种之间没有明显差异。

· 体外稳定荧光肌动蛋白丝的制备。

 

肌动蛋白染色在确定活细胞和固定细胞中细胞骨架的结构和功能方面非常有用。肌动蛋白细胞骨架在活细胞中是非常动态和不稳定的结构，但是在探测或染色肌动蛋白结构之前可以用多聚甲醛固定。 

 

材料

鬼笔环肽是来自蘑菇伞形毒蕈的七氨基酸肽毒素，其特异性地并且以高亲和力（Kd 20nM）结合至肌动蛋白的聚合形式（F-肌动蛋白）。鬼笔环肽将肌动蛋白聚合的临界浓度降低至小于1μg/ ml，从而充当聚合增强剂。鬼笔环肽已经用专有的绿色荧光染料标记，使其可用于染色组织培养细胞和组织切片中的肌动蛋白丝（1，参见图1）和无细胞制剂。Acti-stain™488鬼笔环肽标记的肌动蛋白丝保留了未标记肌动蛋白的许多功能特征，包括它们与肌球蛋白相互作用的能力。Actin-stain TM 488鬼笔环肽以橙色固体形式提供。 

注意：鬼笔环肽是有毒的，必须小心处理（LD50人= 2mg / Kg）。

用抗 – 纽蛋白（红色），Dapi（蓝色细胞核）和F-肌动蛋白染色的Swiss 3T3细胞用Acti-stain TM 488（绿色F-肌动蛋白，Cat。＃PHDG1）染色。

 

Acti-stain TM 488的吸光度和荧光扫描。将标记的鬼笔环肽稀释到甲醇中，并分别在350-750和500-750nm之间扫描其吸光度和激发光谱。吸光度峰值在500nm处，荧光在550nm处。

问题1：我可以使用荧光标记的鬼笔环肽染色活细胞中的F-肌动蛋白吗？

答案1：  不幸的是，不，鬼笔环肽不能用于在活细胞中染色F-肌动蛋白。鬼笔环肽用于在固定细胞中染色F-肌动蛋白。荧光鬼笔环肽仅与F-肌动蛋白的天然四级结构结合，提供低背景。鬼笔环肽结合的正确固定条件是在PBS中的3.7％（v / v）多聚甲醛10分钟，因为它保留了鬼笔环肽的高亲和力结合所必需的季铵蛋白结构。甲醇固定破坏天然构象，因此不适合用鬼笔环肽染色F-肌动蛋白。为了监测活细胞中的肌动蛋白动力学，建议微量注射罗丹明标记的肌动蛋白（Cat。＃APHR或AR05）。有关更多信息，请参阅这些数据表。
