LBIS® 小鼠/大鼠高分子量脂联素 ELISA 试剂盒 LBIS® High Molecular Adiponectin-Mouse/Rat

Adiponectin（脂联素）是脂肪细胞分泌的一种细胞因子。作为脂肪细胞因子，控制脂肪代谢和胰岛素感受性、是抗糖尿病、抗动脉粥样硬化、抗炎症的重要物质。血液中的脂联素通过聚集单聚体形成３聚体、６聚体或者是 12-18 聚体。三聚体（LMW）通过胶原三螺旋链的非共价相互作用以及球状体 C1q 域的疏水相互作用形成。三聚体聚集形成六聚体（MMW）或者更大的多聚体（HMW）。

Adiponectin 与各种各样的生长因子相结合有明显的亲和性，将其隔离能影响细胞的生长、血管新生和细胞组织的重建。血液中 HMW 的测定值除了表示总脂联素以外，同时明确地反映出 BMI 和性别、体重减轻的影响、糖耐量、肝脏的胰岛素感受性、代谢综合征和2型糖尿病。预计 HMW 的测定比起总脂联素的测定，对于代谢综合征和 DM2 的分析更有帮助。

分析原则

Eagle Biosciences Adiponectin ELISA Assay Kit是一种使用两种特异性和高亲和力抗体的所谓三明治分析。样品中的脂连蛋白与涂覆在微量滴定板上的抗体结合。在下一步骤中，第二种特异性抗脂联素 – 抗体依次与固定的脂联素结合。第二抗体是生物素化的，并且将与链霉抗生物素蛋白 – 过氧化物酶 – 酶缀合物一起施用。在封闭的底物反应中，根据样品的脂联素水平定量催化颜色的转变。

测定背景
脂联素是30kDa蛋白质，其血清蛋白质百分比为0.01％。它主要由脂肪细胞合成，但肌细胞和肝细胞也具有合成脂联素的能力。到目前为止，IGF-I是合成中仅有一个已知的天然电感。它由胶原蛋白样N末端和球状C末端结构域组成[1]。体内脂联素出现时具有不同的寡聚体。除三聚体和二聚体外，还存在高分子多聚体[1-3]。到目前为止，已知有两种不同的受体，虽然组织中的分布不同，但这两种受体都是无处不在的表达。脂联素受体1（AdipoR1）特别适用于肌肉和合成肝组织中的AdipoR2 [4]。

直到现在，对人体组织的重要性尚不清楚。项研究表明，脂联素与BMI呈负相关，因此它可能与能量代谢有关，例如通过调节脂肪酸氧化。除了与BMI的相关性外，脂联素水平与胰岛素抵抗相关[5-7]，因此也与II型糖尿病相关。脂联素也与葡萄糖和脂肪代谢相关[8,9]。

此外，它参与炎症过程[11-15]，因此它对于动脉硬化[4,5,16]和冠状动脉[17,18]的出现具有重要意义，因此血浆中脂联素水平的测定可用于估计冠心病的风险[19,20]。除此之外，脂联素影响进一步的生理过程，例如血管生成[21,22]。

标准
