蛋白S是单链维生素K依赖性蛋白,被认为在凝血和补体级联反应中均起作用(1,2)。血浆中循环的大约60%的蛋白S与C4b结合蛋白(C4BP)形成复合物。已经表明,损伤后,蛋白S与带负电荷的磷脂的g-羧基谷氨酸(gla)依赖性结合可能起到将C4BP集中在细胞表面的作用。

在凝血系统中,蛋白质S起到抗凝血辅因子蛋白质的作用。在Ca2 +和磷脂囊泡(Kd = 6×10-9M)存在的情况下,活化的蛋白C(APC)与蛋白S形成1:1的化学计量配合物(3)。在蛋白质S的存在下,血浆和未刺激的血小板表面观察到APC因子Va和VIIIa因子失活的速率有适度增加(3-10倍)。与C4BP结合的蛋白S不具有APC辅因子活性。近,已经描述了增强蛋白S活性的另一种结合蛋白(4)。凝血酶引起的蛋白S的蛋白水解失活已被提议作为该系统中的调节机制。凝血酶的单次切割通过去除含有gla结构域的NH2末端肽(Mr = 8000)消除了蛋白S辅因子的活性。
