内容	BrdU试剂，固定溶液预稀释抗BrdU检测器过氧化物酶山羊抗小鼠IgG 结合物稀释剂底物 50X板洗涤浓缩终止液
应用	BrdU细胞增殖试验

BrdU增殖测定涉及使用细胞层作为固相将BrdU掺入微量滴定板中培养的细胞中。在后2至24小时的培养期间，将BrdU加入微量滴定板的孔中。BrdU将被整合到分裂细胞的DNA中。为了使抗体与掺入的BrdU细胞结合，必须固定，透化并使DNA变性。这一切都是通过使用Fixing Solution进行处理一步完成的。将检测器抗BrdU单克隆抗体移液到孔中并孵育1小时，在此期间它结合任何掺入的BrdU。洗掉未结合的抗体并加入辣根过氧化物酶缀合的山羊抗小鼠抗体，其与检测抗体结合。

的辣根过氧化物酶催化的显色底物四甲基联苯胺（TMB）的从（添加停止试剂后或黄色）的无色溶液至蓝色溶液的转换，其强度正比于结合的BrdU的细胞中的量。使用分光光度计定量有色反应产物。
