炭疽毒素Anthrax Toxins

炭疽杆菌是炭疽病的病原体。炭疽杆菌产生的主要毒力因子是γ连接的聚-D-谷氨酸胶囊和由三种成分组成的外毒素，保护性抗原（PA），致死因子（LF）和水肿因子（EF）。这些蛋白质不是单*毒的，而是以二元组合起作用。PA（细胞结合成分）与LF（酶部分之一）的复合物被称为“致死毒素”并可导致死亡。PA和酶促EF一起引起皮肤水肿。分泌后，PA被膜肽酶切割，使63kDa羧基末端片段寡聚化成七聚体。切割是暴露EF和LF的结合位点的必要步骤。组装的PA复合物和相关的LF或EF通过胞吞作用进入细胞。PA介导LF和EF向细胞质的转移，其中这些酶识别并蛋白水解其靶标。LF组分是一种锌依赖性金属蛋白酶，可裂解丝裂原活化蛋白激酶激酶（MAPKK），从而削弱其功能，从而阻断MAPK信号通路。EF是钙调蛋白依赖性腺苷酸环化酶，其在细胞中产生过量的环AMP，导致水肿。目前的研究集中于开发有效的疫苗以及筛选LF酶活性的有效抑制剂。

本产品是序列中含有PA tag、EGFP和6×His tag的肽，在N末端或C末端分别存在PA tag序列。本产品可用于蛋白质印迹和免疫沉淀的对照。

◆新型亲和标签系统“PA tag”是什么？

PA tag是利用了人平足蛋白的PLAG结构域内序列（GVAMPGAEDDVV）的亲和标签。由于PA tag与抗PA tag抗体的结合力强，特异性高，可以高效地纯化目的蛋白。此外，它可在中性条件下再生抗体结合磁珠，可以循环利用，降低运行成本。

已知PA tag形成蛋白环状结构中的II型的β-转角结构，抗PA tag抗体能与插入环状结构域内的PA tag相结合。为此，PA tag系统拥有环状结构，未来可应用于膜蛋白的纯化和检测。

◆Protocol目的蛋白的纯化

１．哺乳类动物细胞转染(HEK293T)
　　转染处于40-60%融合状态的细胞
　　培养基：D-MEM, 5%FBS

２．回收培养上清和吸附蛋白树脂
　　转染48-72小时后，回收约420 mL培养上清，加入8 mL抗PA标签抗体琼脂糖珠，混合2小时（2-10℃）

３．树脂回收
　　用50 mL的纯化柱。回收步骤2)中的琼脂糖珠

４．树脂洗涤
　　加入珠4倍量的TBS 缓冲液（20 mM Tris-HCl, 150 mM NaCl），清洗5次

５．PA标签肽从树脂中竞争性洗脱
　　加入经1×TBS溶液, pH 7.5溶解过的PA 标签肽（终浓度0.1 mg/mL），体积量需为珠的一倍，

　　然后分10次回收PA标签融合蛋白X

６．SDS-PAGE

　　分别用不同浓度的TBS溶液（0.15 M、0.5 M、1.0 M NaCl）进行调整，用琼脂糖珠50倍量清洗。在回收的各珠中加入0.5 mL 0.1 M Glycine-HCl(pH 2.7)，室温条件下平稳震动5 min。

　　离心后回收上清，用1 M Tris-HCl (pH 9.0)做中和处理。

　　SDS-PAGE后进行银染，比较去除污染蛋白之后，不同浓度下的结果。

MagCapture™ HP抗PA tag抗体磁珠

　　PA tag系统是在日本开发并使用的利用多肽标签（12个氨基酸残基）与大鼠单克隆抗体（NZ-1）结合的亲和纯化标签系统^1）。NZ-1对PA tag具有高亲和性和特异性，通过免疫沉淀可一步纯化出高纯度的PA tag融合蛋白^2）。它还可以应用于蛋白印迹，流式细胞术，免疫细胞染色，利用PA tag洗涤溶液，可以简单地回收使用过的磁珠^1,3）。近年来，通过解析PA tag和NZ-1复合物的晶体结构，明确了PA tag标记β-转角的方法^4）。利用该特点还发表了《将PA tag插入膜蛋白环区中的应用》的应用文章^4）。

　　这一次，FUJIFILM Wako推出了抗PA tag抗体固相化的磁珠。可用于免疫沉淀法中PA tag融合蛋白的纯化和检测。

确认了对K562细胞裂解液中表达的PA tag融合EGFP蛋白的回收性能。结果表明，可在各种洗脱条件下纯化得到高纯度的重组PA tag融合EGFP蛋白。

确认了抗PA tag抗体磁珠可再回收性能。结果表明，即使回收使用三次，也不会降低回收性能，可提取到重组PA tagg融合EGFP蛋白。另外，回收使用了PA tag Washing Solution［产品编号：169-27261］。

PA tag肽竞争性洗脱次数的研讨实验

研讨了是否能通过多次重复竞争性洗脱，提高回收量。结果表明，通过多次重复PA tag的竞争性洗脱，可以提高重组PA tag融合EGFP蛋白的回收量。

亲和纯化标签蛋白！

　　PA tag 系统是利用高亲和性&高特异性单抗（NE-1）识别肽标签（GVAMPGAEDDVV）的重组蛋白新型检测系统。PA tag 系统中的抗体琼脂糖可通过免疫沉淀法纯化 PA tag 融合蛋白，并可在洗涤后获得再生。中性 pH 条件下 PA tag 肽简单方便地竞争性洗脱 PA tag 融合蛋白。

　　PA tag 系统的亲和性和特异性优于现有标签系统，抗体琼脂糖可再生 60 次，极大地降低成本，可用于免疫细胞化学、免疫印迹和流式细胞术，是动物宿主细胞重组蛋白生产的有效工具。

保护性抗原（PA）是炭疽毒素的受体结合成分。原始的83kDa蛋白质被细胞弗林蛋白酶样蛋白酶切割，形成活化的63kDa单体PA63。在这种形式中，PA63与其他PA63分子结合形成环状七聚体。PA63还具有对于与细胞表面上形成的孔相邻的酶LF和EF特异性的结合位点，用于易位。对于醉酒细胞，PA应与EF或LF一起使用约7倍过量。
