什么是外泌体？

OptiPrep™（碘克沙醇

纯化外泌体的宜密度梯度培养基

OptiPrep™是60％碘克沙醇在水中的无菌内毒素测试溶液，密度为1.32 g / ml。

碘克沙醇是作为X射线造影剂开发的，因此已经进行了严格的临床测试。它是非离子性的，对细胞无毒并且在代谢上是惰性的。碘克沙醇溶液可以在所有有用的密度下制成等渗的。碘克沙醇溶液的粘度和重量克分子渗透压浓度低。

Optiprep™使用高分辨率密度梯度超速离心技术进行外来体纯化

高分辨率（12％–36％）碘克沙醇密度梯度分级分离膜封闭的sEV与非囊泡（NV）组分将 OptiPrep™（碘克沙醇）密度介质在冰冷的PBS中立即制备，然后用于产生不连续步骤（12 ％–36％）的渐变。将sEV的粗颗粒（P120）重悬于冰冷的PBS中，并与冰冷的碘克沙醇/ PBS混合，制成终的36％碘克沙醇溶液。将悬浮液添加到离心管的底部，将碘克沙醇浓度递减的PBS溶液小心地铺在顶部，产生完整的梯度。以相同的方式生成没有样品的相同梯度，以用于稍后确定馏分密度。底部负载的12％–36％梯度在120,000 x下进行超速离心

外泌体是脂质双层限定的直径约100nm的囊泡。癌细胞通过分泌促进癌症侵袭和转移的外来体，对周围细胞产生不利影响。我们想知道抑制癌细胞来源的外泌体功能是否会抑制癌症转移。为此，我们使用了与外泌体结合的抗体来抑制源自癌细胞的外泌体的作用。

寻找能识别人乳腺癌细胞外来体的抗体用于具有人乳腺癌细胞的小鼠异种移植模型。首先，我们要检查抗人CD9和抗人CD63是否会结合到从乳腺癌细胞条件培养基分离的外泌体表面。外泌体的小尺寸阻止了常规显微镜的使用。因此，有必要采用免疫电子显微镜。但是，免疫电子显微镜具有挑战性，需要与靶标结合良好的抗体。

抗人CD9（克隆12A12）和抗人CD63（克隆8A12）在免疫沉淀方面具有良好的记录，因此有望在免疫电子显微镜中进行测试。这些试剂很容易检测到CD63。但是，检测CD9之前，需要进行实质性修改，才能成功染色（图1）。另外，出于该实验的目的，重要的是所选抗体对人类而非小鼠具有*的特异性，以防止毒性和脱靶效应。在蛋白质印迹实验中发现该抗体具有非常强的人特异性，可以轻松检测到来自人而非小鼠的CD9和CD63（图2）。

建立了两种抗体的检测方案和人类特异性后，有可能进行体内研究。在乳腺癌的鼠异种移植模型中施用抗人CD9（克隆12A12）和抗人CD63（克隆8A12），以确定是否有可能抑制转移。在此模型中，我们能够成功抑制癌症转移（参考文献1）。我们认为该实验的成功*归因于我们使用的高质量抗体。我希望这些抗体能够帮助更多的人进行研究。

外泌体是直径约100 nm的小细胞外分泌脂质双层分隔的囊泡。ISEV（细胞外囊泡学会）建议使用术语细胞外囊泡（EVs）来指代分泌性囊泡的异质类，其中包括衍生自多囊泡内体的囊泡。近的外泌体研究已经开始阐明外泌体在某些疾病中的作用。但是，要了解外泌体的产生机理，必须具有：1）生理学上了解外泌体的作用；2）了解外泌体引起的疾病。阐明癌细胞外泌体分泌的机制可能导致新的癌症治疗方法。

为了追踪外泌体产生的动力学，我们使用抗人CD63抗体（克隆8A12；可从Cosmo Bio USA获得）进行免疫染色实验。使用免疫荧光显微镜，我们在前列腺癌细胞系PC-3M（图1）和结肠癌细胞系HCT116（图2）中观察到许多CD63阳性颗粒样结构。将来，我们将癌细胞与抗人CD63抗体（克隆8A12）和其他内体标记物抗体共同染色，以探索癌细胞产生外泌体的能力。抗人CD63抗体（克隆8A12）的高特异性使其可以成功用于体内靶向外泌体以抑制癌症转移。

1975年，乔治·科勒（George Kohler）和塞萨尔·米尔斯坦（CésarMilstein）发明了单克隆抗体生产技术，并因此在1984年获得了诺贝尔生理学或医学奖。单克隆抗体*改变了制药业，为新型诊断和靶向疗法打开了大门。它不仅有益于医学实践，也有益于基础研究。并非所有的单克隆抗体都等效。对于每种特定的应用，识别“好”抗体很重要。通常，“好”抗体是识别单个分子并与其分子靶标牢固结合的抗体。

在外泌体研究中获得“良好抗体”尤为重要。外泌体是脂质双层分隔的囊泡，直径约100 nm，在其管腔和膜中含有各种蛋白质。使用免疫电子显微镜，免疫印迹，ELISA，免疫沉淀和免疫亲和柱，已使用抗体来检测和分离体液或培养上清液中的外泌体。这些应用需要“优质”抗体。

由于外泌体是异质的，因此根据其来源，它们可能会展示并包含不同的分子。但是，蛋白质的一个子集很可能在不同类型的外泌体之间共享。目前，好的泛外泌体标记是四跨膜蛋白CD9，CD63和CD81。我的外泌体研究始于构建外泌体检测系统的目标。我努力寻找在我的检测系统中识别外泌体标记的单克隆抗体。尽管我尝试了许多克隆，但直到发现抗CD9（克隆12A12）和抗CD63（克隆8A12）（图1和参考2），我都没有获得好的结果。这些抗体在蛋白质印迹（图2）和免疫电子显微镜中表现良好。这些抗体的人类特异性由其无法检测小鼠CD9和小鼠CD63证实。该特性在小鼠的人异种移植模型系统中很有用，在这种系统中，抗体不会与内源性CD9和CD63反应。如果在检测人外泌体时遇到问题，则抗CD9（克隆12A12）和抗CD63（克隆8A12）抗体可能会有用。

外来体通过ESCRT复合物的作用从多囊泡内体中包含的囊泡中产生。这些腔内囊泡被分泌为直径为50-150nm的细胞外囊泡。近来，随着外泌体包含蛋白质，脂质和核酸的发现，许多注意力集中在它们作为细胞间通讯载体的功能上。

我们正在分析衰老相关的分泌表型，其部分特征是炎症细胞因子和趋化因子的分泌增加。衰老细胞还提高了外泌体的分泌（参考文献1）。我们通过正常人成纤维细胞的连续传代（复制性衰老）和引入激活的癌基因RasV12（癌基因诱导的衰老）来诱导细胞衰老。温育48小时后，通过超速离心回收条件培养基中的外来体。回收的细胞外物质的大小分布通过纳米颗粒跟踪分析进行表征。衰老细胞中分泌的外泌体的数量是年轻细胞的30至50倍（图1，顶部）。为了进一步比较衰老与年轻外来体，我们使用抗人CD81单克隆抗体（克隆12C4，以1：1000稀释度使用；来自Cosmo Bio USA）进行了蛋白质印迹实验。结果表明，从衰老中回收的CD81蛋白量相对于年轻外来体有所增加（图1，底部）。还观察到从衰老细胞中回收的外泌体对MCF-7乳腺癌细胞具有促进生长的作用（参考文献2）。

外泌体的生理作用和诊断/治疗潜力仍未*了解。我们正在分析正常细胞和衰老细胞分泌外泌体的详细分子机制，以期阐明外泌体的生物学意义。此外，通过研究衰老

EXOTRAP™外来体分离离心柱套件

实验发现

外泌体是膜包裹的30至100 nm的囊泡，内含蛋白质和核酸，并由各种细胞分泌。通过获取循环的通路，它们可以将信号传输到远处的小区。已经观察到，通过外来体的细胞间通讯与抗原呈递，细胞凋亡，炎症，肿瘤进展和恶性转化有关。预计外泌体将在各个领域中发挥重要作用，例如在药物输送系统的开发中。

我们对病毒感染的细胞与附近未感染的细胞之间的相互作用感兴趣。当病毒感染的细胞与未感染的细胞混合培养后，荧光免疫染色可以检测未感染细胞中的病毒蛋白，呈点状信号。从这个发现，我对作为细胞间通讯介体的外泌体产生了兴趣。

由于涉及冗长而困难的方法（传统上包括超速离心），因此比较从各种来源和在不同条件下回收的外泌体具有挑战性。因此，从操作简便（任何人都可以纯化），纯化时间（可以在短时间内完成纯化）和应用的多功能性（可以分析外泌体本身以及所含蛋白质和核酸）的角度出发，我们进行了商业化探索可用的外泌体纯化试剂盒，发现CosmoBio USA的“ ExoTrap™外泌体分离旋转柱试剂盒”是理想的纯化解决方案。

ExoTrap™的优势在于，可以在仅30分钟的“短时间内”从少量样品中纯化外泌体。作为外泌体研究的初学者，ExoTrap™使我能够通过蛋白质印迹法检测与在纯化前培养上清液中无法检测到的外泌体相关的病毒蛋白。而且，我能够证明在从外泌体抑制剂（GW4869）处理过的细胞的培养上清液中回收的ExoTrap™纯化的外泌体中未检测到病毒蛋白（1）。尽管ExoTrap™可能不适合廉价地纯化大量外泌体，但强烈建议从小体积快速简便地纯化外泌体。
