描述

通过CD133免疫荧光染色鉴定从包括Her-2阳性癌症患者的癌症患者的外周血，腹水或胸腔积液富集的循环肿瘤干细胞（CTSC）。可以根据需要同时进行与针对Her-2和上皮细胞等附加标志物的抗体的共染色。

使用该试剂盒在液相中对细胞进行免疫荧光染色很容易，快速且适合于CTC的基因组分析。

风湿病、组织纤维化、癌症研究用

结缔组织生长因子CTGF（Connective Tissue Growth Factor）是脐静脉和血管内皮细胞等中产生的一种分泌蛋白，分子量约为38kDa。CTGF由Module1-4共四个部分组成，控制着软骨细胞的增殖分化和细胞粘附。有报告显示，CTGF与癌症、肾功能衰竭、肝功能衰竭、风湿等各种疾病也有所关联。

本产品是抗CTGF小鼠单克隆抗体，能够识别通过DNA免疫法建立的Native form的CTGF，具有中和活性。

◆使用示例（in vitro中和实验）破骨细胞的分化诱导抑制

实验条件

1. 从人全血中提取PBMCs，分离CD14⁺细胞。

2. 将CD14⁺细胞在α-MEM（添加10% FBS）、M-CSF（25 ng/mL）、sRANKL（40 ng/mL）中培养7天（37℃, 5% CO₂），使其向破骨细胞分

化。

3. 破骨细胞进行TRAP染色后，在光学显微镜下统计TRAP阳性细胞数。

结果

无论哪一种CTGF抗体都抑制了CTGF对破骨细胞的分化诱导。

◆使用示例（in vivo中和实验）风湿病模型动物（小鼠）的关节炎抑制

关节炎指标标准

POINT	观察结果
0	无变化
1	肢体局部发红。又或是1根手指的红肿。
2	手、脚关节肿胀。又或是2根以上的手指肿胀。
3	手、脚底有中度~明显的肿胀。
4	肢体明显的红肿、硬化。

实验条件

1. 向DBA/1J (Female, 7-8weeks) 小鼠的尾部皮内给药由胶原蛋白二型(TypeⅡ collagen) 和弗氏完全佐剂 (CFA)等量混合而成的乳状液

（100 µL/小鼠），诱发关节炎。

2. 在免疫发生前一周开始腹腔注射抗CTGF抗体（10 mg/kg），每周1次，共8次。

3. 每周对四肢关节炎进行1次评价，直至免疫发生的第8周（评价标准参考上述表格）。

结果

通过给药抗CTGF module1抗体，风湿病模型小鼠的关节炎受到了抑制。

《科学》：如何让癌症干细胞正常

来自加拿大麦克马斯特大学等处的研究人员发表文章，发现一种药物：硫利达嗪（Thioridazine）能成功消灭人体癌症干细胞，从而避免了传统癌症治疗方法引起的毒副作用。这一重要的成果公布在5月24日的Cell杂志上。

领导这一研究的是加拿大麦克马斯特大学干细胞与癌症研究院科学主任Mick Bhatia教授，他表示，“这项研究发现不同寻常之处在于，这种已有药物实际杀死癌症干细胞的方式——将这些癌症干细胞变成正常非癌变细胞”。Bhatia教授一直致力于癌症干细胞的研究，他曾*次论证了人类正常干细胞和癌症干细胞的区别。

15年以来，科学家们都认为干细胞是许多癌症的源头。1997年加拿大的科学家*次在某种白血病中识别出了癌症干细胞，自此癌症干细胞便陆续在血癌，乳腺癌，脑癌，肺癌，胃肠道癌症，前列腺癌以及卵巢癌中被发现。

与正常干细胞不同，癌症干细胞不会分化成稳定的，非分裂细胞类型，之前Bhatia教授曾利用一种筛选平台，按照其表达的基因和起反应的药物来定义这两者之间的差别，从而能够开发能更好地对付癌细胞而不损害健康细胞的疗法和药物。

而这项研究中，Bhatia教授和他的同事通过药物研发模型，筛选了上百种化合物，从中鉴别出来将近20种潜在的癌症干细胞特异性药物，其中一种可能zui有潜力的药物就是这种称为硫利达嗪的化合物。

硫利达嗪（Thioridazine）实际上是一种治疗精神疾病的药物，可以通过靶向大脑中的多巴胺受体，来治疗精神躯体障碍所致焦虑和紧张状态。这种药物其实并不能杀死癌症干细胞，但是却能促进癌症干细胞分化，从而消耗干细胞的自我更新能力。

研究人员发现，硫利达嗪能由此消灭白血病干细胞，并且不会影响正常血液干细胞，将白血病患者体内的癌症干细胞蛋白与正常血细胞中的蛋白进行比较，能帮助解释这种特异性。血癌细胞与正常血液干细胞不同，前者表面能表达一种多巴胺受体，而多巴胺受体也能在某些乳腺癌细胞中表达。

“这进行了一些解释”，Bhatia教授说，而且这也说明多巴胺受体也许能作为一种罕见肿瘤驱动细胞的生物标记物。

利用这些发现，Bhatia教授研究组目前正计划进行硫利达嗪这种FDA批准药物的临床实验，结合标准的成人急性髓细胞白血病抗癌药物，以及这种药物进行治疗。

“我们很高兴看到能将这种药物引入临床”，Bhatia教授说，“我们也希望这一平台能成为其它癌症干细胞药物筛选的流水线。”

IN SITE™ Metastasis Kit

IN SITE™ Metastasis Kit是由猪骨、肺和肝脏来源的组织特异性细胞外基质（Tissue / Organ-specific ECM）组合而成的试剂盒，可用于研究转移性癌症及其治疗药的研发。这些组织特异性细胞外基质保留了组织/器官ECM的原始微环境因子，并因为可作为体外疾病模型制备的基质而备受关注^1,2)。

通过使用该试剂盒作为癌细胞培养基质，可以在体外重现癌症转移多发部位（骨、肝、肺）的微环境，从而能够构建更合适的体外肿瘤模型。构建的体外肿瘤模型可用于癌症治疗药物的筛选测试等，还能提高癌症治疗药物研发的成功率。

◆组织特异性ECM在体外肿瘤模型中的重要性

合适的体外肿瘤模型，是细胞抗癌药物药效评估和筛选测试的关键。迄今已有多种体外模型，但多数由于与实际患者的肿瘤组织和性质差异较大，导致体外模型与体内的药效不同，这也成为了抗癌药物研发的一大难题。

◆应用案例2：癌转移部位及药物敏感性研究

癌细胞在不同的转移部位具有不同的药物敏感性。

在TissueSpec™ 骨/肝/肺 ECM 水凝胶或塑料表面上接种乳腺癌细胞（a）BT-549和（b）T47-D，添加抗癌药（Paclitaxel：5 μM）或Vehicle（DMSO），48 h后，通过检测OD值观察各条件下细胞的生存率。
